Uses of Vardif - What is Vardenafil Used For?

Vardif Dosage - How Much Vardenafil Should I Take?

Vardenafil Hydrochloride tablets (Vardif) is taken orally, approximately 60 minutes before sexual activity. Vardif may be taken with or without food. The maximum recommended dosing frequency is one tablet per day (one tablet in any 24 hour period). The dosage of Vardif tablets (Vardenafil) is given below.

Vardenafil is taken as needed: For most patients, the starting dose is 10 mg, up to once daily. Increase to 20 mg or decrease to 5 mg based on efficacy/tolerability.
A starting dose of 5 mg Vardenafil Hydrochloride tablets (Vardif) should be considered in patients 65 years of age or more.
In case undergoing treatment with potent or moderate inhibitors of CYP3A4, the dose of Vardif should be adjusted as follows:

• Ritonavir: No more than 2.5 mg in a 72-hour period
• Indinavir, Saquinavir, Atazanavir, Ketoconazole 400 mg daily, Itraconazole 400 mg daily, Clarithromycin: No more than 2.5 mg in a 24-hour period
• Ketoconazole 200 mg daily, Itraconazole 200 mg daily, Erythromycin: No more than 5 mg in a 24-hour period.

In patients on stable alpha-blocker therapy the recommended starting dose of Vardenafil tablets is 5 mg. The recommended starting dose of Vardenafil Hydrochloride tablets is 5 mg in patients with moderate hepatic impairment (Child-Pugh B). The maximum dose in patients with moderate hepatic impairment must not exceed 10 mg in any 24 hour period.

Vardif Storage

Store Vardif tablets at controlled room temperature 25°C (77°F). Excursions permitted from 15°C to 30°C (59°F to 86°F). Protect from light.

Vardif Contraindications

Vardif tablets are contraindicated in patients with a hypersensitivity to Vardenafil Hydrochloride or any inactive ingredient present in tablets. Vardif are also contraindicated in the following cases:
Administration of Vardif tablets with nitrates (either regularly and/or intermittently) and nitric oxide donors is contraindicated. Consistent with the effects of PDE5 inhibition on the nitric oxide/cyclic guanosine monophosphate pathway, PDE5 inhibitors including Vardenafil Hydrochloride could potentiate the hypotensive effects of nitrates.

Concomitant use of Vardif (Vardenafil tablets) with Indinavir, Ritonavir, Ketoconazole, or Itraconazole is contraindicated, as they are potent inhibitors of CYP3A4.

Vardif (Vardenafil tablets) is contraindicated in patients with erectile dysfunction with a previous episode of non-arteritic anterior ischaemic optic neuropathy (NAION)

The co-administration of PDE5 inhibitors, including Vardif tablets, with guanylate cyclase stimulators, such as Riociguat, is contraindicated as it may lead to potentially life-threatening episodes of symptomatic hypotension or syncope.

Warnings and Precautions

Below are the warnings and precautions to be taken when using Generic Vardenafil Hydrochloride tablets (Vardif)

• Cardiovascular Effects: Patients should not use Vardif if sex is inadvisable due to cardiovascular status.
• Risk of Priapism: In the event that an erection lasts more than 4 hours, the patient should seek immediate medical assistance.
• Effects on the Eye: Patients should stop use of Vardif, and seek medical attention in the event of sudden loss of vision in one or both eyes, which could be a sign of nonarteritic anterior ischemic optic neuropathy (NAION). Vardif should be used with caution, and only when the anticipated benefits outweigh the risks, in patients with a history of NAION. Patients with a "crowded" optic disc may also be at an increased risk of NAION.
• Sudden Hearing Loss: Patients should stop Vardif and seek medical attention in the event of sudden decrease or loss in hearing.
• Alpha-Blockers: Caution is advised when PDE5 inhibitors are coadministered with alpha-blockers. In some patients, concomitant use of these two medicine classes can lower blood pressure significantly leading to symptomatic hypotension (for example, fainting).
• QT Prolongation: Patients with congenital QT syndrome or taking class IA or III antiarrhythmics should avoid using Vardif tablets. Low levels of Potassium or Magnesium in the blood could also increase the risk of QT prolongation.
• This medicine may make the patient dizzy or cause vision changes. Alcohol could increase the level of dizziness. Do not drive, use machinery, or do anything that needs alertness or clear vision until you can do it safely. Limit alcoholic beverages.
• Vardenafil Hydrochloride tablets (Vardif) have not been administered to patients with bleeding disorders or significant active peptic ulceration. Therefore Vardif tablets must be administered to these patients after careful benefit-risk assessment.
• Treatment for erectile dysfunction should generally be used with caution in patients with anatomical deformation of the penis (such as angulation, cavernosal fibrosis or Peyronie's disease)or in patients who have conditions that may predispose them to priapism (such as sickle cell anemia, multiple myeloma, or leukemia).
• Sudden decrease or loss of hearing has been reported in a few postmarketing and clinical trial cases with the use of PDE5 inhibitors, including Vardenafil tablets. These events, which may be accompanied by tinnitus and dizziness, have been reported in temporal association to the intake of PDE5 inhibitors, including Vardenafil tablets.
• Use of Vardenafil Tablets (Vardif) in patients with known hereditary degenerative retinal disorders such as retinitis pigmentosa is not recommended until information is available regarding efficacy and safety of this medicine when administered to such patients.

Vardenafil With Alcohol

When Alcohol (0.5 g/kg body weight: approximately 40 mL of absolute alcohol in a 70 kg person) and Vardenafil 20 mg when dosed simultaneously, plasma levels were not altered . Vardenafil 20 mg tablets did not potentiate the hypotensive effects of alcohol during the 4-hour observation period in healthy volunteers when administered with alcohol (0.5 g/kg body weight).

Vardenafil Overdosage

The maximum dose of Vardenafil Tablets (Vardif) for which human data are available is a single 120 mg dose administered to eight healthy male volunteers. The majority of these subjects experienced reversible back pain/myalgia and/or "abnormal vision."

In cases of Vardif overdose, standard supportive measures should be taken as required. Renal dialysis is not expected to accelerate clearance because vardenafil is highly bound to plasma proteins and is not significantly eliminated in the urine.

Vardenafil During Pregnancy
