Uses of Crestor (Rosuvastatin Calcium Tablets)
Crestor (Rosuvastatin Calcium Tablets) is used in combination with exercise, diet, and weight-loss to treat high cholesterol and related conditions and to prevent cardiovascular disease when the response to diet and exercise is not adequate.
It is also indicated for risk reduction of MI, stroke, and arterial revascularization procedures in patients without clinically evident CHD, but with multiple risk factors. It also lowers the risk of heart attacks and strokes. Crestor is used to treat the following indications:
- adult patients with primary hyperlipidemia and mixed dyslipidemia along with diet to reduce elevated total-C, LDL-C, ApoB, nonHDL-C, and TG levels and to increase HDL-C.
- pediatric patients 8 to 17 years of age with heterozygous familial hypercholesterolemia (HeFH) to reduce elevated total-C, LDL-C and ApoB after failing an adequate trial of diet therapy.
- pediatric patients 7 to 17 years of age with homozygous familial hypercholesterolemia (HoFH) to reduce LDL-C, total-C, nonHDL-C and ApoB as an adjunct to diet, either alone or along with other lipid-lowering treatments.
- adult patients with hypertriglyceridemia along with a proper diet
- adult patients with primary dysbetalipoproteinemia (Type III hyperlipoproteinemia) as a supplement to the diet
- adult patients with homozygous familial hypercholesterolemia (HoFH) to reduce LDL-C, total-C, and ApoB
- slowing the progression of atherosclerosis as part of a treatment strategy to lower total-C and LDL-C as a supplement to the diet

The approximate elimination half-life of Rosuvastatin Calcium is 19 hours and its time to peak plasma concentration is reached in 3 to 5 hours following oral administration.
Crestor (Rosuvastatin Calcium Tablets) - Dosage
The dose range for Crestor (Rosuvastatin Calcium Tablets) in adults is 5 mg to 40 mg orally once daily. The normal starting dose is 10 to 20 mg once daily. The usual starting dose in adult patients with homozygous familial hypercholesterolemia is 20 mg once daily.
The maximum dose of 40 mg should be used only for those patients who have not achieved their LDL-C goal utilizing the 20 mg dose.
Crestor can be administered as a single dose at any time of day, with or without food. The tablet must be swallowed whole.
Pediatric Dosing:
In heterozygous familial hypercholesterolemia, the recommended dose range is 5 to 10 mg orally once daily in patients 8 to less than 10 years of age, and 5 to 20 mg orally once daily in patients 10 to 17 years of age. In homozygous familial hypercholesterolemia, the recommended dose is 20 mg orally once daily in patients 7 to 17 years of age.
Dosing in Asian Patients:
In Asian patients, consider starting therapy with 5 mg once daily due to increased Rosuvastatin plasma concentrations. The increased systemic exposure must be taken into consideration when treating Asian patients not adequately controlled at doses up to 20 mg/day.
Dosage adjustments are required in patients undergoing Concomitant Therapy and those with Severe Renal Impairment.
Crestor Side Effects
Most frequent Crestor (Rosuvastatin Calcium Tablets) side effects with an incidence rate of 2% or more are a headache, myalgia, abdominal pain, asthenia, and nausea. Stop taking Crestor Tablets and contact the prescribing doctor immediately if any of the following rare but serious side effects occur:
- muscle pain, tenderness, or weakness
- fever
- jaundice: yellowing of the skin or eyes
- dark colored, or foamy urine
- muscle cramps or spasms
- muscle pain, stiffness, tenderness, wasting, or weakness
- Abdominal pain or stomach pain
- extreme tiredness or weakness
- clay-colored stools
- skin rash
- unpleasant breath odor
- vomiting of blood

Crestor Tablets Contraindications
Crestor tablets are contraindicated in patients with a hypersensitivity to Rosuvastatin Calcium or any inactive ingredient present in tablets. Hypersensitivity reactions including rash, pruritus, urticaria, and angioedema have been reported with this medicine. Crestor tablets are also contraindicated in the following cases:
Patients with active liver disease, which may include unexplained persistent elevations of hepatic transaminase levels
Lactation. Limited data indicate that Rosuvastatin Calcium is present in human milk.
Pregnancy
Warnings and Precautions
The following are the precautions to be taken when undergoing therapy with Crestor tablets:
Skeletal Muscle Effects: Cases of myopathy and rhabdomyolysis with acute renal failure secondary to myoglobinuria have been reported with HMG-CoA reductase inhibitors, including Crestor tablets. These risks could occur at any dose level but are increased at the highest dose (40 mg).
Liver Enzyme Abnormalities: It is recommended that liver enzyme tests be performed before starting therapy with Crestor tablets and if signs or symptoms of liver injury occur.
There have been rare postmarketing reports of fatal and non-fatal hepatic failure in patients taking statins, including Rosuvastatin Calcium tablets. If serious liver injury with clinical symptoms and/or hyperbilirubinemia or jaundice occurs during treatment with Crestor tablets, promptly stop therapy. If an alternate etiology is not found, do not restart treatment with Crestor.
Concomitant Coumarin Anticoagulants: Caution should be exercised when anticoagulants are given in conjunction with Crestor tablets because of its potentiation of the effect of coumarin-type anticoagulants in prolonging the prothrombin time/INR.
Proteinuria and Hematuria: In the Crestor tablets clinical trial program, dipstick-positive proteinuria and microscopic hematuria were seen among Crestor tablets treated patients. These findings were more frequent in patients taking Crestor 40 mg, when compared to lower doses of Crestor tablets.
Endocrine Effects: Increases in HbA1c and fasting serum glucose levels have been reported with HMG-CoA reductase inhibitors, including Crestor tablets. Based on clinical trial data with Crestor tablets, in some instances these increases may exceed the threshold for the diagnosis of diabetes mellitus.
Crestor (Rosuvastatin Calcium Tablets) Overdosage
There is no specific treatment in the case of Crestor overdose. In the event of overdose, the patient must be treated symptomatically and supportive measures instituted as required. Hemodialysis does not significantly enhance clearance of Rosuvastatin.
Rosuvastatin Calcium During Pregnancy
Rosuvastatin Calcium (Crestor tablets) has been classified by the US FDA as Pregnancy Category X. If this medication is used during pregnancy, or if the patient becomes pregnant while taking this drug, the patient should be informed of the potential harm to the fetus.
Animal studies have failed to reveal evidence of teratogenicity, however, studies conducted in rats demonstrated decreased pup survival, decreased fetal body weight among female pups, and delayed ossification. Do not use this medicine without your doctor's consent in case you are pregnant. Stop using this medication and inform your doctor immediately if you become pregnant during treatment.
Nursing Considerations
This medicine must not be used during breastfeeding due to the potential for serious adverse events in nursing infants and the concern over disruption of infant lipid metabolism.
